本計畫測試、應用及改良移動式藻類及毒素監測系統,針對國內14座離島水庫,應用分子生物、光學、及免疫學技術,定期、快速監測主要水庫水質及藻華、毒素、大腸菌相關資訊,以保障民眾用水安全。
計畫中首先收集國內外相關文獻,了解目前國內外相關移動式實驗室監測項目與方式。並改良現有監測系統,包括針對微囊藻及魚腥藻兩種目標藻改用雙重基因即時定量測試、提升柱孢藻目標基因之定量及監測專一性、增加蛤蚌毒素測試等,且均已成功應用於現地測試。
本年度共進行8座本島、6座離島水庫、35次採樣、116個現場樣品分析。檢測結果顯示,5個本島水庫樣品中,微囊藻毒素或柱孢藻毒素有超過0.2 μg/L的情形,所幸所有後續採樣清水樣品均遠低於建議值,顯示供水仍是安全。在臭味物質部份,2座水庫,霉味物質MIB超過國外飲用水建議值(10 ng/L)。在6座離島水庫採樣中,發現所有水庫均有藻類代謝物問題,4座水庫微囊藻毒素及柱孢藻毒素均超過0.2 μg/L,1座水庫兩者甚至超過1 μg/L,所有水庫霉味物質MIB均超過10 ng/L。
本計畫採樣中,對於取水口毒素及毒藻基因濃度超過建議數值(0.2 μg/L)者,現場均立即增採下游淨水廠原水及清水樣品,並快速將結果通知水利署及水庫管理單位,以提供水庫及水廠管理單位參考與應變。計畫並初步完成8座水庫水質特性資訊收集、藻毒事件文宣製作、以及網站架設。在教育訓練部份,本計畫於金門太湖水廠進行離島藻華應變演練,模擬藻華事件發生時,水廠及相關單位因應、通報、及解決方法;並於成功大學舉辦教育訓練,除藻類毒素及有害微生物基本知識外,並加入離島水庫情境模擬範例,以及現場實作教學,提升水庫及水廠人員藻毒應變與處理相關知識。
英文摘要
The aim of this project is to test, apply, and modify a mobile laboratory for the quick monitoring of toxigenic Microcystis and Cylindrospermopsis, cyanotoxins, geosmin-producers, earthy-musty odorants, and Total Coliform in 14 reservoirs in Taiwan. Three key instruments, an in-vivo fluorescence (IVF) probe, a quantitative polymerase chain reaction (qPCR) machine, and an enzyme-linked immunosorbent assay (ELISA) reader, were installed within the mobile laboratory, together with a microscope, a spectrometer, and an anemometer.
The information about the development of mobile laboratory and device was first collected, showing that most available technologies are relevant to the measurement of conventional water quality parameters. In this study, a duplex PCR for both microcystin genes and geosmin genes, pks genes for cylindrospermopsin, and saxitoxi, were integrated into the existing analytical platform. The modified methods were successfully applied in the field this year.
The mobile laboratory was successfully tested on-site for 35 times in 10 reservoirs for the analysis of 116 samples in Taiwan. Nine reservoirs studied were found to have the concentrations of either microcystins or cynlindrospermopsin concentration higher than 0.2 μg/L, and 8 reservoirs had earthy odorant MIB concentration higher than the suggested drinking water quality guideline value (10 ng/L).
The water quality information were collected for 8 major reservoirs in Taiwan. In addition, to promote the knowledge relevant to cyanotoxins, information about occurence, treatment and response for cyanotoxins in drinking water systems was also integrated in a flyer and a website. An emergency response drill was designed by the project team and was held in Tai-Hu Waterworks, Kinmen for the training of reservoir and waterwork managers and operators. To provide knowledge about treatment and response of cyanotaxins for the operators in reservoirs and water utilities, a training workshop was also held at National Cheng Kung University.
